Histologic Studies in Changes of Skeletal Muscle after Compression of Arteries and Release from Compression in Limb of Rabbit

Abstract
In order to study histopathologic changes and histochmical changes of glycogen and connective tissue distribution in the muscle fibers of ischemic skeletal muscle, one of the hind limbs of rabbits was applied with elastic rubber tourniquet to obliterate arterial circulation of limbs. The author observed histopathologic changes and histochemical changes of glycogen and connective tissue distribution in the gastrocnemius, soleus and anterior tibialis at the interval of 1st, 2nd, 4th, 8th, 12th, 24th and 48th hours after application of the tourniquet, and in order to study regeneration of degenerated and necrotic muscle fibers, at the 24th and 48th hours after release of the tourniquet which had been applied for 12 hours. The muscle was stained with H-E, PAS, Masson Trichrom and Van Gieson stainings.
The results obtained were as follows:
1. From one hour to two hours after the application of elastic rubber tourniquet, muscle fibers was little difference comparsion with normal groups. From four hours, swelling, atropy and hyaline degeneration of muscle fibers was evident. From twentyfour hours, rupture, hemorrhage and sporadic necrosis of muscle fibers was evident.
2. From four hours after application of elastic rubber tourniquet, amount of connective tissue in muscle fibers increased, and markedly increased from twenty-four hours. Glycogen in muscle fibers is completely disappered after four hours.
3. At twenty-four hours after release of elastic rubber tourniquet which had been applied twelve hours, hemorrhage and infiltration of polymorphonuclear leucocyte in muscle fibers were markedly increased. At forty-eight hour, appearance of muscle fibers begin to change the normal shape.
4. From twenty-four hours to forty-eight hours after release of elastic rubber tourniquet which had been applied twelve hours, connective tissue distributon in muscle fibers were similar, but generally more increase rather than muscle fibers which applied tourniquet. Glycogen distribution in muscle fibers was reappeared with positive reaction for PAS stainings at twenty-four hours and forty-eight hours.
